Taxol Induces bcl-2 Phosphorylation and Death of Prostate Cancer Cells1
نویسندگان
چکیده
Treatment of prostate cancer cell lines expressing bcl-2 with taxol induces bcl-2 phosphorylation and programmed cell death, whereas treat ment of ¿>c/-2-negativeprostate cancer cells with taxol does not induce apoptosis. bcl-2 phosphorylation seems to inhibit its binding to box since less box was observed in immunocomplex with bcl-2 in taxol-treated cancer cells. These findings support the use of the anticancer drug taxol for the treatment of ftc/-2-positive prostate cancers and other ec/-2-positive malignancies, such as follicular lymphoma.

متن کاملEpigallocatechin-3-Gallate Induces Apoptosis through Up-regulation of Bax and Down-regulation of Bcl-2 in Prostate Cancer Cell Line
Background and Aims: Epigallocatechin-3-gallate (EGCG) is a polyphenolic compound from green tea, which its anticancer effects on many types of cancers have been confirmed, but the molecular mechanism by which EGCG induces apoptosis remains unknown. The aim of the present study was to investigate anti-proliferative properties and apoptotic signaling pathway of EGCG on PC3 human prostate cancer ...
متن کاملTaxol induces apoptosis in cortical neurons by a mechanism independent of Bcl-2 phosphorylation.
Bcl-2, an antiapoptotic protein, protects cells against many but not all forms of apoptosis. For example, Bcl-2 does not protect non-neuronal cells against taxol, a microtubule-stabilizing agent. The underlying mechanism for the ineffectiveness of Bcl-2 against taxol has been the subject of intense interest. Data from non-neuronal cells indicate that taxol-induced apoptosis requires activation ...
متن کاملInduction of Apoptosis by ß-Lapachonein Human Prostate Cancer Cells1
ß-Lapachone, a plant product, has been shown to be a novel inhibitor of DNA topoisomerase I, with a mode of action different from camptothecin and a chemical structure distinct from those of current anti-cancer drugs. We observed that ß-lapachone, at concentrations of less than 8 UM, induces cell death with characteristics of apoptosis in human prostate cancer cell lines. This effect of ß-la...
متن کاملTaxol-induced apoptosis and phosphorylation of Bcl-2 protein involves c-Raf-1 and represents a novel c-Raf-1 signal transduction pathway.
c-Raf-1 (Raf-1) is a central component of signal transduction pathways stimulated by various growth factors, protein kinase C, and other protein kinases. Raf-1 activation is thought to be initiated at the plasma membrane after its recruitment by Ras. Raf-1 activation is associated primarily with proliferation and cell survival, but it has also been implicated in apoptosis. Raf-1 has also been s...
